Optimisation of activity by the application of structure-activity relationships. Quantitative structure-activity relationships, Ferguson's principle. Linear free-energy relationships and Hansch analysis. Synthesis of drug substances including problems with scaling up to manufacture, impurity profiles and how these are controlled. Mechanisms of drug degradation. Introduction to nuclear magnetic resonance spectroscopy, mass-spectrometry. Pro-drugs as a method of drug design and delivery, examples include optimisation of antibiotic activity, miscellaneous drugs and tissue targeting. Structure activity relationships and receptor interaction. Benzodiazepines. Isotopes as tracer compounds. Vitamins. Steroids. Prostaglandins. Analysis of drugs and Peptide drugs as therapeutic agents.
